Concept:
The "Bulbosum method" in barley involves crossing common barley (Hordeum vulgare) with Hordeum bulbosum. After fertilization, the H. bulbosum chromosomes are eliminated, leaving a haploid embryo.
Step 1: Hybridization Prep.
First, to prevent self-pollination in the female parent, Emasculation (C) is performed. This is followed by the actual crossing through Pollen collection and cross pollination (A) using the second species.
Step 2: Rescue and Doubling.
Because these hybrid embryos often abort on the plant due to chromosome elimination, they must be saved via Embryo culture (B) in a lab. Once a haploid plantlet is formed, its fertility is restored via Colchicine treatment (D) to double the chromosomes.
